Can someone point is to a practical application of "Replenishment Class".  We do not know how to get set up our Replenishment Classes.  Any help would be greatly appreciated.

One for Purchase and one for Transfer, that's usually what I see.

Then you use the Replenishment Classes on the Stock Item setup to decide whether Replenishment will create a line on the Create Purchase Orders (PO505000) screen or create a line on the Create Transfer Orders (SO509000) screen when replenishment determines that the Stock Item is needed.
